چکیده

Alpha-synuclein (-synuclein) is considered a key player in Parkinson's disease (PD), but the exact relationship between-synuclein aggregation and dopaminergic (DA) neurodegeneration remains unresolved. There is increasing evidence that neuroinflammatory processes are closely linked to DA cell death, but whether the inflammatory process is causally involved in PD or rather reflects secondary consequences of nigrostriatal pathway injury is still under debate. We evaluated the therapeutic effect of the immunophilin ligand FK506 in a rAAV2/7-synuclein overexpression rat model. Treatment with FK506 significantly increased the survival of dopaminergic neurons in a dose-dependent manner. No reduction in-synuclein aggregation was apparent in this time window, but FK506 significantly lowered the infiltration of both T helper and cytotoxic T cells and the number and subtype of microglia/ macrophages. These data suggest that the anti-inflammatory properties of FK506 decrease neurodegeneration in this-synuclein-based PD model, pointing to a causal role of neuroinflammation in the pathogenesis of PD.

The Endotoxin-Induced Neuroinflammation Model of Parkinson's Disease

Parkinson's disease (PD) is a common neurodegenerative disorder characterized by the progressive loss of dopaminergic (DA) neurons in the substantia nigra. Although the exact cause of the dopaminergic neurodegeneration remains elusive, recent postmortem and experimental studies have revealed an essential role for neuroinflammation that is initiated and driven by activated microglial and infiltr...

Protective effect of chlorogenic acid in an experimental model of Parkinson’s disease induced by 6-hydroxydopamine in rats

Introdution: Parkinson's disease is the second most common neurodegenerative disease. Considering the antioxidant and neuroprotective properties of chlorogenic acid, the purpose of this study was to evaluate the neuroprotective effect of this substance in an experimental model of Parkinson's disease. Methods: In this experimental study, Wisar male rats (n = 32) were divided into 4 groups: sham...

Neuroprotection and restoration of the nigrostriatal dopaminergic system in 6-OHDA lesioned rat model of Parkinson's disease: Role of GDNF and TGF expressing Zuckerkandl's organ

Zuckerkandl’s organ (ZK) is an extra adrenal para-ganglion and has the ability to express glial cell line derived neurotrophic factor (GDNF) and transforming growth factor (TGF). It is also a source of dopamine and norepinephrine. In the present study, the neuroprotective and restorative potential of ZK was studied by transplanting it into the striatum of adult rats either before or after the i...

Cyclooxygenase and Neuroinflammation in Parkinson’s Disease Neurodegeneration

Cyclooxygenase (COX) expression in the brain is associated with pro-inflammatory activities, which are instrumental in neurodegenerative processes such as Parkinson's disease (PD). It is discussed that drugs with the capacity to rescue dopaminergic neurons from microglia toxicity and neuroinflammatory processes may result in an amelioration of parkinsonian symptoms by delaying the onset or slow...
